Biography

Dr. Bones obtained his BSc in Analytical Science (Chemistry) and PhD in Analytical Chemistry from Dublin City University (DCU) under the guidance of Prof. Brett Paull. Following completion of his doctoral studies, Jonathan took a position at the Center for Bioanalytical Science in DCU, focused on the development and application of long monolithic silica columns for high efficiency LC-MS profiling of cell culture media, a collaboration with Bristol Myers Squibb.

Jonathan joined Prof. Pauline M. Rudds group at NIBRT in 2007 based at the UCD Conway Institute of Biomolecular and Biomedical Research, representing NIBRT in the EU FP6 GLYFDIS – ‘Glycans in Bodily Fluids, Potential for Disease Diagnostics’ Consortium before becoming the GlycoAnalytical Team Leader within the Dublin-Oxford Glycobiology Laboratory.

In 2010 Jonathan was appointed as the ‘John Hatsopoulos Research Scholarship’ at the Barnett Institute of Chemical and Biological Analysis at Northeastern University in Boston, MA, working under the mentorship of Prof. Barry L. Karger investigating new approaches for glycomics and the application of proteomics for a better understanding of mammalian cell bioprocessing.

In 2012, Dr Bones returned to NIBRT following receipt of an SFI Starting Investigator Research Grant and founded his Characterisation and Comparability Laboratory which focuses on the development and application of advanced liquid phase separations coupled to high resolution MS for the complete characterisation of bioprocesses and expressed therapeutic glycoproteins. The group consists of 18 team members supported through funding from SFI, EI, FP7 as well as direct industry support.

His research activities have been internationally recognised through his inclusion on the recently announced Medicine Maker Power List of the top 100 most influential scientists in the field of biopharmaceutical manufacturing and analysis.

Research Areas

Dr. Bones research focuses on the development and application of liquid phase separations and mass spectrometry (LC-MS and CE-MS) for the analysis of complex biological systems. Dr. Bones current research program is funded through receipt of awards from Science Foundation Ireland and Industrial Collaborators.

Current research areas include:

(1) Application of quantitative proteomics, metabolomics and lipidomics to understand how alterations in bioprocess conditions yield molecular manifestations in CHO cells that result in altered product quality;

(2) Development of advanced LC-MS based platforms for quantitative glycomics with applications in bioprocessing and clinical biomarker discovery;

(3) Multidimensional separations coupled to mass spectrometry for complex bioanalysis; and

(4) Development of analytical platforms to support process optimization and PAT.

Thermofisher Scientific

ThermoFisher Scientific and Dr. Jonathan Bones collaborate on the development of analytical solutions for the characterisation of complex biopharmaceuticals.

This collaboration sees NIBRT develop workflows on the Thermo Scientific biomolecule column range with its associated consumable portfolio in conjunction with sophisticated Thermo Scientific liquid chromatography systems and advanced Thermo Scientific Orbitrap high resolution mass spectrometers.
